This missense variant replaces methionine with arginine at codon 10 of the RAD51C protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with ovarian cancer (PMID: 21750962). This variant has been identified in 2/251448 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.M10R variant (also known as c.29T>G), located in coding exon 1 of the RAD51C gene, results from a T to G substitution at nucleotide position 29. The methionine at codon 10 is replaced by arginine, an amino acid with similar properties. This alteration was identified in 1/232 ovarian cancer patients not selected for family history (Vuorela M et al. Breast Cancer Res. Treat. 2011 Dec;130:1003-10). This alteration was also detected in 1/5589 German BRCA1/2-negative probands with breast cancer (Hauke J et al. Cancer Med, 2018 Apr;7:1349-1358). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces methionine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 10 of the RAD51C protein (p.Met10Arg). This variant is present in population databases (rs730881936, gnomAD 0.002%). This missense change has been observed in individual(s) with ovarian cancer (PMID: 21750962). ClinVar contains an entry for this variant (Variation ID: 182841). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change does not substantially affect RAD51C function (PMID: 37253112). RNA analysis performed to evaluate the impact of this missense change on mRNA splicing indicates it does not significantly alter splicing (internal data). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
