This missense variant replaces aspartic acid with asparagine at codon 1138 of the BRIP1 protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with BRIP1-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The c.3411_3412delTGinsCA variant (also known as p.D1138N), located in coding exon 19 of the BRIP1 gene, results from an in-frame deletion of TG and insertion of CA at nucleotide positions 3411 to 3412. This results in the substitution of the aspartic acid residue for an asparagine residue at codon 1138, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
