Genes affected
BRIP1 (HGNC:20473): (BRCA1 interacting helicase 1) The protein encoded by this gene is a member of the RecQ DEAH helicase family and interacts with the BRCT repeats of breast cancer, type 1 (BRCA1). The bound complex is important in the normal double-strand break repair function of breast cancer, type 1 (BRCA1). This gene may be a target of germline cancer-inducing mutations. Uncertain significance, criteria provided, single submitterclinical testingGeneDxNov 25, 2023Not observed at significant frequency in large population cohorts (gnomAD); In silico analysis supports that this missense variant does not alter protein structure/function; This variant is associated with the following publications: (PMID: 28678401, 29636988, 11301010, 27978560) -
Uncertain significance, criteria provided, single submitterclinical testingQuest Diagnostics Nichols Institute San Juan CapistranoJul 20, 2023In the published literature, this variant has been reported in an individual with head and neck squamous cell carcinoma, in an individual with colon cancer, in individuals with breast cancer and in an unaffected control (PMIDs: 27978560 (2016), 28678401 (2017), and 33471991 (2021)). The frequency of this variant in the general population, 0.000031 (4/129142 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant. -
Hereditary cancer-predisposing syndrome Uncertain:2
Uncertain significance, criteria provided, single submitterclinical testingColor Diagnostics, LLC DBA Color HealthNov 01, 2022This missense variant replaces arginine with histidine at codon 1035 of the BRIP1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in one individual with colorectal cancer (PMID: 27978560). In a large breast cancer case-control study, this variant was identified in 2/60464 cases and 1/53460 controls (OR=1.768, 95%CI 0.16 to 19.503; p-value=1 (Leiden Open Variation Database DB-ID BRIP1_000314) (PMID: 33471991). This variant has been identified in 5/282814 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
Uncertain significance, criteria provided, single submitterclinical testingAmbry GeneticsSep 14, 2023The p.R1035H variant (also known as c.3104G>A), located in coding exon 19 of the BRIP1 gene, results from a G to A substitution at nucleotide position 3104. The arginine at codon 1035 is replaced by histidine, an amino acid with highly similar properties. This alteration was detected in 1/450 probands from a cohort of individuals with early-onset colorectal cancer referred for multi-gene panel testing (Pearlman R et al. JAMA Oncol 2017 Apr;3(4):464-471). This alteration was also detected in an individual diagnosed with head and neck squamous cell carcinoma (Chandrasekharappa SC et al. Cancer, 2017 Oct;123:3943-3954). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

Uncertain significance, criteria provided, single submitterclinical testingWomen's Health and Genetics/Laboratory Corporation of America, LabCorpAug 04, 2022Variant summary: BRIP1 c.3104G>A (p.Arg1035His) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 251452 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3104G>A has been reported in the literature in an individual affected with Colon Cancer (Pearlman_2017). This report does not provide an unequivocal conclusion about association of the variant with Hereditary Breast and Ovarian Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Six ClinVar submitters (evaluation after 2014) cite the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -
Familial cancer of breast;C1836860:Fanconi anemia complementation group J Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpFeb 01, 2024This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 1035 of the BRIP1 protein (p.Arg1035His). This variant is present in population databases (rs367816363, gnomAD 0.004%). This missense change has been observed in individual(s) with colon cancer (PMID: 27978560). ClinVar contains an entry for this variant (Variation ID: 128183). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
