The p.D773N variant (also known as c.2317G>A), located in coding exon 15 of the BRIP1 gene, results from a G to A substitution at nucleotide position 2317. The aspartic acid at codon 773 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 773 of the BRIP1 protein (p.Asp773Asn). This variant is present in population databases (rs146091205, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with BRIP1-related conditions. ClinVar contains an entry for this variant (Variation ID: 583239).
