The c.484C>A variant (also known as p.R162R), located in coding exon 4 of the BRIP1 gene, results from a C to A substitution at nucleotide position 484. This nucleotide substitution does not change the amino acid at codon 162. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site; however, direct evidence is insufficient at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear. -
