This missense change has been observed in individual(s) with autosomal dominant paramyotonia congenita (PMID: 32594687). In at least one individual the variant was observed to be de novo. This sequence change replaces arginine, which is basic and polar, with glycine, which is neutral and non-polar, at codon 1448 of the SCN4A protein (p.Arg1448Gly). This variant is not present in population databases (gnomAD no frequency). ClinVar contains an entry for this variant (Variation ID: 1451612). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant disrupts the p.Arg1448 amino acid residue in SCN4A. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 7676326, 8619545). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
