The p.S626R variant (also known as c.1878T>A), located in coding exon 6 of the AXIN2 gene, results from a T to A substitution at nucleotide position 1878. The serine at codon 626 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
Oligodontia-cancer predisposition syndrome Uncertain:1
This sequence change replaces serine, which is neutral and polar, with arginine, which is basic and polar, at codon 626 of the AXIN2 protein (p.Ser626Arg). This variant is present in population databases (rs767756290, gnomAD 0.0009%). This missense change has been observed in individual(s) with neuroblastoma (PMID: 27009842). ClinVar contains an entry for this variant (Variation ID: 533225). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt AXIN2 protein function with a positive predictive value of 80%.
