The highest population minor allele frequency in gnomAD v2.1.1 is 0.00004 in African population, which is lower than the ClinGen ACADVL Variant Curation Expert Panel threshold (<0.001) for PM2_Supporting, meeting this criterion (PM2_Supporting). At least one patient with this variant displayed enzyme levels of ≤20% of normal, which is highly specific for very long-chain acyl-CoA dehydrogenase (VLCAD) deficiency (PP4_moderate, PMID: 17999356). This variant has been reported in additional individuals with VLCAD deficiency, however this information is insufficient to use toward classification (PMIDs: 31497477, 27246109, 9973285, 10384387). This variant has been observed with the variant [c.1813C>T, p.Arg613Trp] which is classified as likely pathogenic by the ClinGen ACADVL Variant Curation Expert Panel(ClinVar SCV) in an individual with VLCAD deficiency however the phase of these variants was not confirmed (PMID: 17999356, 0.25 PM3 points).
