The p.E440Q variant (also known as c.1318G>C), located in coding exon 9 of the DNAI2 gene, results from a G to C substitution at nucleotide position 1318. The glutamic acid at codon 440 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This sequence change replaces glutamic acid, which is acidic and polar, with glutamine, which is neutral and polar, at codon 440 of the DNAI2 protein (p.Glu440Gln). This variant is present in population databases (rs182986650, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with DNAI2-related conditions. ClinVar contains an entry for this variant (Variation ID: 241447). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
