UNC13D (HGNC:23147): (unc-13 homolog D) This gene encodes a protein that is a member of the UNC13 family, containing similar domain structure as other family members but lacking an N-terminal phorbol ester-binding C1 domain present in other Munc13 proteins. The protein appears to play a role in vesicle maturation during exocytosis and is involved in regulation of cytolytic granules secretion. Mutations in this gene are associated with familial hemophagocytic lymphohistiocytosis type 3, a genetically heterogeneous, rare autosomal recessive disorder. [provided by RefSeq, Jul 2008]

This frequency is not significantly higher than estimated for a pathogenic variant in UNC13D causing Familial Hemophagocytic Lymphohistiocytosis (0.00052 vs 0.0027), allowing no conclusion about variant significance. c.3160A>G has been reported in the literature as a double heterozygous genotype in combination with a missense variant c.272C>T (p.A91V) in the PRF1 gene in at-least one individual with Familial Hemophagocytic Lymphohistiocytosis (FHL) (Zhang_2014). The authors proposed a model of synergistic heterozygosity in FHL between two genes involved in cytotoxic lymphocyte degranulation. This report does not provide unequivocal conclusions about association of the variant with Familial Hemophagocytic Lymphohistiocytosis.
