Its main function is to catalyze the synthesis of palmitate from acetyl-CoA and malonyl-CoA, in the presence of NADPH, into long-chain saturated fatty acids. In some cancer cell lines, this protein has been found to be fused with estrogen receptor-alpha (ER-alpha), in which the N-terminus of FAS is fused in-frame with the C-terminus of ER-alpha. The c.7516G>A (p.V2506M) alteration is located in exon 43 (coding exon 42) of the FASN gene. This alteration results from a G to A substitution at nucleotide position 7516, causing the valine (V) at amino acid position 2506 to be replaced by a methionine (M). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
