Experimental functional studies using heterozygous patient-derived peripheral lymphocytes have shown that this variant causes significant decrease in LDLR expression levels and LDL binding activity (PMID: 12522687). This variant is found to be absent in the general population database, gnomAD and interpreted as likely pathogenic/pathogenic by several ClinVar submitters including the ClinGen Familial Hypercholesterolemia Variant Curation Expert Panel (ClinVar ID: 3744).
