This missense change has been observed in individuals with clinical features of cerebral arteriopathy with subcortical infarcts and leukoencephalopathy and lateral meningocele syndrome (PMID: 12146805, 12395806, 15229130; Invitae). This sequence change replaces cysteine, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 76 of the NOTCH3 protein (p.Cys76Arg).
