This sequence change replaces histidine with glutamine at codon 407 of the DNAAF3 protein (p.His407Gln). The histidine residue is weakly conserved and there is a small physicochemical difference between histidine and glutamine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with DNAAF3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The glutamine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. The p.H407Q variant (also known as c.1221C>A), located in coding exon 9 of the DNAAF3 gene, results from a C to A substitution at nucleotide position 1221. The histidine at codon 407 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved.
