GALNT3 (HGNC:4125): (polypeptide N-acetylgalactosaminyltransferase 3) This gene encodes UDP-GalNAc transferase 3, a member of the GalNAc-transferases family. This family transfers an N-acetyl galactosamine to the hydroxyl group of a serine or threonine residue in the first step of O-linked oligosaccharide biosynthesis. Individual GalNAc-transferases have distinct activities and initiation of O-glycosylation is regulated by a repertoire of GalNAc-transferases. The protein encoded by this gene is highly homologous to other family members, however the enzymes have different substrate specificities. [provided by RefSeq, Jul 2008]

ClinVar contains an entry for this variant (Variation ID: 7793). This variant has been observed in individual(s) with familial tumoral calcinosis (PMID: 15133511). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is present in population databases (rs375879489, gnomAD 0.02%). This sequence change falls in intron 8 of the GALNT3 gene. It does not directly change the encoded amino acid sequence of the GALNT3 protein. It affects a nucleotide within the consensus splice site. -
