ClinVar contains an entry for this variant (Variation ID: 206829). This variant is also known as IVS23+1A>G. This variant has been observed in individual(s) with clinical features of SCN1A-related conditions (PMID: 21248271, 23195492, 29655203, 33278787; Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change falls in intron 23 of the SCN1A gene. It does not directly change the encoded amino acid sequence of the SCN1A protein. It affects a nucleotide within the consensus splice site. -

c.4476+1 A>G: IVS23+1 A>G in intron 23 of the SCN1A gene (NM_001165963.1) The c.4476+1 A>G splice site mutation in the SCN1A gene has been reported previously in two individuals with classic Dravet syndrome (Zuberi et al., 2011), and as a de novo mutation in an individual with severe myoclonic epilepsy of infancy (Wang et al., 2012). This mutation destroys the canonical splice donor site in intron 23, and is expected to cause abnormal gene splicing. The variant is found in EPILEPSY panel(s). -
