The TTN c.107123C>T variant is predicted to result in the amino acid substitution p.Pro35708Leu. This variant has been reported in an individual with dilated cardiomyopathy (Table S3, Mazzarotto et al. 2020. PubMed ID: 31983221). This variant is reported in one allele out of ~246,000 alleles in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -

This sequence change replaces proline with leucine at codon 35708 of the TTN protein (p.Pro35708Leu).  There is a moderate physicochemical difference between proline and leucine. The frequency data for this variant (rs71423567) in the population databases is unreliable, as metrics indicate poor quality at this position in the ExAC database. This variant has not been reported in the literature in individuals with a TTN-related disease.
