The p.H1269R variant (also known as c.3806A>G), located in coding exon 48 of the COL3A1 gene, results from an A to G substitution at nucleotide position 3806. The histidine at codon 1269 is replaced by arginine, an amino acid with highly similar properties. Based on data from ExAC, the G allele has an overall frequency of less than 0.01% (1/105450). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Ehlers-Danlos syndrome, type 4 Uncertain:1
This sequence change replaces histidine, which is basic and polar, with arginine, which is basic and polar, at codon 1269 of the COL3A1 protein (p.His1269Arg). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
