This sequence change replaces alanine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 219 of the MATN3 protein (p.Ala219Asp). This variant is present in population databases (rs28939677, gnomAD 0.007%). This missense change has been observed in individual(s) with multiple epiphyseal dysplasia (PMID: 14729835, 21922596; Invitae). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 7543). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt MATN3 protein function with a positive predictive value of 80%.
