BMPR2 (HGNC:1078): (bone morphogenetic protein receptor type 2) This gene encodes a member of the bone morphogenetic protein (BMP) receptor family of transmembrane serine/threonine kinases. The ligands of this receptor are members of the TGF-beta superfamily. BMPs are involved in endochondral bone formation and embryogenesis. These proteins transduce their signals through the formation of heteromeric complexes of two different types of serine (threonine) kinase receptors: type I receptors of about 50-55 kD and type II receptors of about 70-80 kD. Mutations in this gene have been associated with primary pulmonary hypertension, both familial and fenfluramine-associated, and with pulmonary venoocclusive disease. [provided by RefSeq, May 2020]

Likely pathogenic, criteria provided, single submitter | clinical testing | ARUP Laboratories, Molecular Genetics and Genomics, ARUP Laboratories | Sep 04, 2019 | The BMPR2 c.353G>A; p.Cys118Tyr variant (rs1085307216) is reported in the literature in several individuals affected with pulmonary arterial hypertension (PAH) (Machado 2006, Pfarr 2011). This variant is absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. The cysteine at codon 118 is moderately conserved, and computational analyses (SIFT, PolyPhen-2) predict that this variant is deleterious. Other amino acid substitutions at this codon and the adjacent cysteine (p.Cys117Tyr, p.Cys118Trp, p.Cys118Ser) have been reported in individuals with PAH and are considered disease-causing (Barozzi 2019, Machado 2006). Further, functional assays indicate that several of these other substitutions (p.Cys117Tyr and p.Cys118Trp) are mislocalized in the cell and exhibit signaling defects (Rudarakanchana 2002, Sobolewski 2008).
