Crystallins constitute the major proteins of vertebrate eye lens and maintain the transparency and refractive index of the lens. This gene and several family members are present in a gene cluster on chromosome 2. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15"). This variant has been reported in the literature in an individual with uni-lateral sporadic congenital cataract (PMID: 27307692). This variant is present in population databases (rs200572745, ExAC 0.01%). This sequence change replaces glutamic acid with lysine at codon 94 of the CRYGC protein (p.Glu94Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. -
