Variant summary
Our verdict is Uncertain significance. The variant received 0 ACMG points: 0P and 0B.
The NM_001244008.2(KIF1A):c.5087G>A(p.Arg1696His) variant causes a missense change involving the alteration of a conserved nucleotide. The variant allele was found at a frequency of 0.0000781 in 1,612,412 control chromosomes in the GnomAD database, with no homozygous occurrence. Variant has been reported in ClinVar as Conflicting classifications of pathogenicity (no stars). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. R1696S) has been classified as Likely benign.

Inborn genetic diseases Uncertain:1
The p.R1595H variant (also known as c.4784G>A), located in coding exon 44 of the KIF1A gene, results from a G to A substitution at nucleotide position 4784. The arginine at codon 1595 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
