Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 9.1e-06 in 220772 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.497T>A in individuals affected with Primary Hyperoxaluria Type 1 and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance. - |

To our knowledge, this variant has not been reported in the literature. A different amino acid substitution at this position (p.Leu166Pro) was reported in a patient with primary hyperoxaluria and functional analysis suggests the p.Leu166 residue is important for protein function (Williams et al 2007. PubMed ID: 17495019; Lage MD et al 2014. PubMed ID: 24718375). This variant is reported in 0.0020% of alleles in individuals of European (non-Finnish) descent in gnomAD. Although we suspect this variant could be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. - |
