Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00039 in 152912 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for a pathogenic variant in AGXT causing Primary Hyperoxaluria Type 1 (0.00039 vs 0.0024), allowing no conclusion about variant significance. c.865C>T has been reported in the literature in individuals affected with Primary Hyperoxaluria Type 1 (example: Rinalt_1999 and Frishberg_2005). These report(s) do not provide unequivocal conclusions about association of the variant with Primary Hyperoxaluria Type 1. At least one publication reports experimental evidence evaluating an impact on protein function. These results showed no damaging effect of this variant (example: Pittman_2012).
