When cloned, it was discovered to be a human homolog of the E. coli mismatch repair gene mutS, consistent with the characteristic alterations in microsatellite sequences (RER+ phenotype) found in HNPCC. This sequence change replaces methionine with isoleucine at codon 67 of the MSH2 protein (p.Met67Ile). The methionine residue is weakly conserved and there is a small physicochemical difference between methionine and isoleucine. -
