This alteration (referred to as IVS8+1G>T) has been reported in two French HNPCC families (Parc Y et al. J. Med. Genet., 2003 Mar;40:208-13) as well as in individuals whose Lynch syndrome-related tumors demonstrated high microsatellite instability (MSI-H) and loss of MSH2 by IHC analysis (van der Post RS et al. J Med Genet, 2010 Jul;47:464-70). (Overbeek LI et al. Br J Cancer, 2007 May;96:1605-12). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In silico splice site analysis predicts that this alteration will weaken the native splice donor site. In addition to the clinical data presented in the literature, alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is classified as a disease-causing mutation. - |

This variant has been reported in multiple individuals and families affected with Lynch syndrome and Lynch syndrome-associated cancers (PMID: 12624141, 17453009, 20591884, 21642682, 26300997, 30723297, 31118792, 30877237). In several of these individuals, tumors displayed loss of MSH2 protein expression via immunohistochemistry analysis and high microsatellite instability (PMID: 17453009, 20591884, 30877237). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of MSH2 function is a known mechanism of disease (clinicalgenome.org).
