ClinVar contains an entry for this variant (Variation ID: 649648). This missense change has been observed in individual(s) with MSH6-related conditions (PMID: 30217226). The p.F432C variant (also known as c.1295T>G), located in coding exon 4 of the MSH6 gene, results from a T to G substitution at nucleotide position 1295. The phenylalanine at codon 432 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. In addition, the CoDP in silico tool predicts this alteration to have a major impact on molecular function, with a score of 0.999 (Terui H et al. J. Biomed. Sci. 2013 Apr;20:25). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
