The p.I1115T variant (also known as c.3344T>C), located in coding exon 5 of the MSH6 gene, results from a T to C substitution at nucleotide position 3344. The isoleucine at codon 1115 is replaced by threonine, an amino acid with similar properties. This alteration was reported in an individual with colorectal cancer (Giráldez MD et al. Clin. Cancer Res., 2010 Nov;16:5402-13). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces isoleucine with threonine at codon 1115 of the MSH6 protein. Computational prediction tools and conservation analyses are inconclusive regarding the impact of this variant on protein structure and function. Splice site prediction tools suggest that this variant may not impact RNA splicing. This variant has been reported in one individual affected with colorectal cancer (PMID: 20924129). This variant has been identified in 1/251372 chromosomes in the general population by the Genome Aggregation Database (gnomAD).
