The enzyme cleaves the distal alpha-1,2-linked glucose residue from the Glc(3)-Man(9)-GlcNAc(2) oligosaccharide precursor. This protein is located in the lumen of the endoplasmic reticulum. Defects in this gene are a cause of type IIb congenital disorder of glycosylation (CDGIIb). Two transcript variants encoding different isoforms have been found for this gene. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This sequence change results in a premature translational stop signal in the MOGS gene (p.Gln800*). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 38 amino acids of the MOGS protein. This variant disrupts a region of the protein in which other variant(s) (p.Thr802Ile) have been observed in individuals with MOGS-related conditions (PMID: 29235540). This suggests that this may be a clinically significant region of the MOGS protein. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
