Variant summary: The CHEK2 c.1114T>A (p.Ser372Thr) variant involves the alteration of a conserved nucleotide, resulting in a missense change in the protein kinase domain (InterPro). 3/4 in silico tools predict a damaging outcome for this variant (SNPsandGO not captured due to low reliability index). This variant is absent in 118554 control chromosomes (ExAC). The variant of interest has not, to our knowledge, been reported in affected individuals via publications and/or reputable databases/clinical diagnostic laboratories, nor evaluated for functional impact by in vivo/vitro studies. The p.S372T variant (also known as c.1114T>A), located in coding exon 10 of the CHEK2 gene, results from a T to A substitution at nucleotide position 1114. The serine at codon 372 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
