The p.L303* pathogenic mutation (also known as c.908T>A), located in coding exon 7 of the CHEK2 gene, results from a T to A substitution at nucleotide position 908. This changes the amino acid from a leucine to a stop codon within coding exon 7. This mutation was identified in a breast cancer cell line derived from an American patient diagnosed with breast cancer at 39 years of age (Wasielewski M et al. Breast Cancer Res. Treat. 2009 Jan; 113(2):285-91). Since premature stop codons are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294). -
