The c.3535A>G (p.N1179D) alteration is located in exon 35 (coding exon 34) of the FANCD2 gene. This alteration results from a A to G substitution at nucleotide position 3535, causing the asparagine (N) at amino acid position 1179 to be replaced by an aspartic acid (D). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.

This sequence change replaces asparagine, which is neutral and polar, with aspartic acid, which is acidic and polar, at codon 1179 of the FANCD2 protein (p.Asn1179Asp). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with FANCD2-related conditions.
