Comput ational prediction tools and conservation analyses suggest that this variant may not impact the protein, though this information is not predictive enough to rul e out pathogenicity. The arginine (Arg) at position 431 is not conserved in mamm als or evolutionary distant species, raising the possibility that a change at th is position may be tolerated. In summary, while the clinical significance of the p.Arg431Cys variant is uncertain, these data suggest that it is more likely to be benign. - |
