The p.R954C variant (also known as c.2860C>T), located in coding exon 15 of the MYLK gene, results from a C to T substitution at nucleotide position 2860. The arginine at codon 954 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant (referred to as p.R885C) has been reported in an individual in a thoracic aortic aneurysm and dissection cohort, but clinical details were limited (Chen ZR et al. J Thorac Dis, 2021 Jul;13:4008-4022). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
