The encoded protein is important for damage sensing and DNA binding, and shows a preference for single-stranded DNA. Mutations in this gene or some other NER components can result in Xeroderma pigmentosum, a rare autosomal recessive disorder characterized by increased sensitivity to sunlight with the development of carcinomas at an early age. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product. This variant is present in population databases (no rsID available, gnomAD 0.0009%). Disruption of this splice site has been observed in individual(s) with xeroderma pigmentosum (PMID: 16081512, 17079196). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as Intron 5.1 A>G at -2. ClinVar contains an entry for this variant (Variation ID: 550100). Studies have shown that disruption of this splice site alters XPC gene expression (PMID: 16081512). Studies have shown that disruption of this splice site results in 83 bp insertion of intron 5 and introduces a premature termination codon (PMID: 16081512). The resulting mRNA is expected to undergo nonsense-mediated decay. For these reasons, this variant has been classified as Pathogenic. - |
