BTD (HGNC:1122): (biotinidase) The protein encoded by this gene functions to recycle protein-bound biotin by cleaving biocytin (biotin-epsilon-lysine), a normal product of carboxylase degradation, resulting in regeneration of free biotin. The encoded protein has also been shown to have biotinyl transferase activity. Mutations in this gene are associated with biotinidase deficiency. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has been observed to be in combination with another BTD variant in an individual affected with biotinidase deficiency (Invitae). ClinVar contains an entry for this variant (Variation ID: 439037). This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with alanine at codon 111 of the BTD protein (p.Pro111Ala). The proline residue is highly conserved and there is a small physicochemical difference between proline and alanine. -
