The MLH1 p.Ser193= variant was not identified in the literature. The variant was identified in dbSNP (ID: rs587781038) as "With Likely benign allele", ClinVar (classified as benign by Invitae and GeneDx; as likely benign by Ambry Genetics, Counsyl and Color), and in UMD-LSDB (3x as neutral). In UMD the variant was identified with a co-occurring likely pathogenic/pathogenic MSH2 variant (c.IVS2+1G>A (c.366+1G>A)), increasing the likelihood that the p.Ser193= variant does not have clinical significance. The variant was not identified in the following control databases: the Exome Aggregation Consortium (August 8th 2016), or the Genome Aggregation Database (Feb 27, 2017). The p.Ser193= variant is not expected to have clinical significance because it does not result in a change of amino acid and is not located in a known consensus splice site. In addition, in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer) do not predict a difference in splicing.
