This variant has not been reported in the literature in individuals affected with MLH1-related conditions. ClinVar contains an entry for this variant (Variation ID: 479670). The p.Q197* pathogenic mutation (also known as c.589C>T), located in coding exon 8 of the MLH1 gene, results from a C to T substitution at nucleotide position 589. This changes the amino acid from a glutamine to a stop codon within coding exon 8. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
