Brugada syndrome Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Invitae | Feb 06, 2022 | This sequence change replaces arginine, which is basic and polar, with lysine, which is basic and polar, at codon 1935 of the SCN10A protein (p.Arg1935Lys). This variant is present in population databases (rs764945982, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with SCN10A-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SCN10A protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |
Cardiovascular phenotype Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Dec 22, 2020 | The p.R1935K variant (also known as c.5804G>A), located in coding exon 27 of the SCN10A gene, results from a G to A substitution at nucleotide position 5804. The arginine at codon 1935 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
