COL7A1 (HGNC:2214): (collagen type VII alpha 1 chain) This gene encodes the alpha chain of type VII collagen. The type VII collagen fibril, composed of three identical alpha collagen chains, is restricted to the basement zone beneath stratified squamous epithelia. It functions as an anchoring fibril between the external epithelia and the underlying stroma. Mutations in this gene are associated with all forms of dystrophic epidermolysis bullosa. In the absence of mutations, however, an acquired form of this disease can result from an autoimmune response made to type VII collagen. [provided by RefSeq, Jul 2008]

Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID:21448560, 9668111,19726672). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt COL7A1 protein function. This variant has been observed in individual(s) with clinical features of autosomal dominant epidermolysis bullosa dystrophica (PMID: 16971478). This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with valine at codon 2015 of the COL7A1 protein (p.Gly2015Val). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and valine. -
