Caveolin proteins are proposed to be scaffolding proteins for organizing and concentrating certain caveolin-interacting molecules. Mutations identified in this gene lead to interference with protein oligomerization or intra-cellular routing, disrupting caveolae formation and resulting in Limb-Girdle muscular dystrophy type-1C (LGMD-1C), hyperCKemia or rippling muscle disease (RMD). Alternative splicing has been identified for this locus, with inclusion or exclusion of a differentially spliced intron. The observed variant frequency is approximately 13.18 fold of the estimated maximal expected allele frequency for a pathogenic variant in CAV3 causing Long QT Syndrome phenotype (3.3e-06), strongly suggesting that the variant is benign. c.115-13G>C has been reported in the literature in individuals affected with DCM without strong evidence for causality (Pugh_2014). These report(s) do not provide unequivocal conclusions about association of the variant with Long QT Syndrome.
