The ANK2 c.5570T>A variant is predicted to result in the amino acid substitution p.Val1857Glu. This variant was reported in four siblings with cardiac arrhythmia (Asadi et al. 2016. PubMed ID: 27110552). This variant is reported in 0.011% of alleles in individuals of European (Non-Finnish) descent in gnomAD (http://gnomad.broadinstitute.org/variant/4-114275344-T-A). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
