Hypertrophic cardiomyopathy Uncertain:2
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Mar 14, 2022 | In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). ClinVar contains an entry for this variant (Variation ID: 45781). This missense change has been observed in individual(s) with hypertrophic cardiomyopathy (PMID: 27532257, 31513939). This variant is present in population databases (rs76757102, gnomAD 0.004%). This sequence change replaces glutamine, which is neutral and polar, with arginine, which is basic and polar, at codon 10 of the MYOZ2 protein (p.Gln10Arg). - |

not specified Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine | Nov 09, 2012 | The Gln10Arg variant in MYOZ2 has not been reported in the literature nor previo usly identified by our laboratory or in large and broad European American and Af rican American populations by the NHLBI Exome Sequencing Project (http://evs.gs. washington.edu/EVS/). This low frequency is insufficient to determine its clinic al significance. Computational analyses (biochemical amino acid properties, cons ervation, AlignGVGD, PolyPhen2, and SIFT) do not provide strong support for or a gainst an impact to the protein. In summary, additional information is needed to fully assess the clinical significance of the Gln10Arg variant. - |

Cardiovascular phenotype Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Sep 29, 2017 | The p.Q10R variant (also known as c.29A>G), located in coding exon 1 of the MYOZ2 gene, results from an A to G substitution at nucleotide position 29. The glutamine at codon 10 is replaced by arginine, an amino acid with highly similar properties. This alteration was detected in a hypertrophic cardiomyopathy genetic testing cohort; however, clinical details were limited (Walsh R et al. Genet. Med., 2017 Feb;19:192-203). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
