not specified Uncertain:1
The p.V260G variant (also known as c.779T>G), located in coding exon 6 of the PDLIM3 gene, results from a T to G substitution at nucleotide position 779. The valine at codon 260 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Primary dilated cardiomyopathy;C0007194:Hypertrophic cardiomyopathy Uncertain:1
This sequence change replaces valine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 260 of the PDLIM3 protein (p.Val260Gly). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with PDLIM3-related conditions. ClinVar contains an entry for this variant (Variation ID: 525142). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt PDLIM3 protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
