Mutations in this gene, as well as in a neighboring gene that lies in a head-to-head configuration, cause Ellis-van Creveld syndrome, an autosomal recessive skeletal dysplasia that is also known as chondroectodermal dysplasia. Mutations in this gene also cause acrofacial dysostosis Weyers type, also referred to as Curry-Hall syndrome, a disease that combines limb and facial abnormalities. The G296A variant in the EVC2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G296A variant is observed in 60/11406 (0.53%) alleles from individuals of Latino background, in the ExAC dataset (Lek et al., 2016). The G296A variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is conserved in mammals. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret G296A as a variant of uncertain significance. -
