This novel 2-base pair deletion in exon 12 of the APC gene (c.1605_1606delTG) causes a frameshift resulting in a premature stop codon (p.Cys535Serfs*3). It was identified in a proband affected with familial adenomatous polyposis (FAP) and desmoid tumor. Segregation analysis confirmed the variant in her two affected children (one daughter and one son), while her healthy daughter did not carry the mutation, supporting co-segregation with disease. -
