The p.P2522T variant (also known as c.7564C>A), located in coding exon 15 of the APC gene, results from a C to A substitution at nucleotide position 7564. The proline at codon 2522 is replaced by threonine, an amino acid with highly similar properties. This alteration was seen in 1/150 unselected patients with recurrent or metastatic prostate cancer. (Isaacsson Velho P et al. Prostate. 2018 04;78:401-407). This amino acid position is well conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
