This alteration has been observed in one individual from a cohort of patients with Cowden/Cowden-like (CS/CS-like) and Bannayan-Riley-Ruvalcaba syndromes (BRRS) without PTEN mutations (Yehia L et al. In addition, in silico predictors for this gene do not accurately predict pathogenicity. This variant has not, to our knowledge, been published in the literature as a pathogenic or benign germline variant. APC Ile2738Val was not observed in large population cohorts (Lek 2016). This variant is located in the EB1 binding domain (Azzopardi 2008). In silico analysis, which includes protein predictors and evolutionary conservation, supports that this variant does not alter protein structure/function. Based on currently available evidence, it is unclear whether APC Ile2738Val is a pathogenic or benign variant. We consider it to be a variant of uncertain significance. - |
