Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Oct 12, 2023 | The p.I1293V variant (also known as c.3877A>G), located in coding exon 25 of the RAD50 gene, results from an A to G substitution at nucleotide position 3877. The isoleucine at codon 1293 is replaced by valine, an amino acid with highly similar properties. In one study, this variant was reported in 1/60,466 breast cancer cases as well as 1/53,461 controls (Dorling et al. N Engl J Med. 2021 02;384:428-439). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
