This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 525 of the SDHA protein (p.Val525Met). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with SDHA-related conditions. ClinVar contains an entry for this variant (Variation ID: 472342). The p.V525M variant (also known as c.1573G>A), located in coding exon 12 of the SDHA gene, results from a G to A substitution at nucleotide position 1573. The valine at codon 525 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
